Identification of a new stripe rust resistance gene YrTZH in Chinese wheat landrace

Abstract

Stripe rust is a major threat to wheat production globally. Breeding disease-resistant cultivars is the best way to control the disease. Chinese wheat landrace Tianzhenhong (TZH) displayed a high level of stripe rust resistance against prevalent Puccinia striiformisf. sp.tritici(Pst) races in China. TZH was crossed with the susceptible landrace Xiangxianhuoshao (XXHS) and subsequent filial generations were obtained. Monogenic segregation was observed in a mapping population TX-87–1 developed by selfing a segregating line from F2:3families of  XXHS/TZH, and the dominant resistance locus was tentatively designated YRTZH. A combination of bulked segregant analysis (BSA) with wheat 55K single nucleotide polymorphism (SNP) array was used to identify flanking markers linked to YRTZH. The YRTZHlocus was mapped to chromosome 1BL and flanked by KASP-TZH-6 and KASP-TZH-11within a 0.8 cM genetic interval corresponding to 20.7 Mb physical region (343.2–363.9 Mb) in the Chinese Spring genome.YrTZHis likely different from previously stripe rust resistance genes/quantitative trait loci on chromosome 1BL. The gene and associated flanking KASP markers could be useful for breeding wheat cultivars with resistance to stripe rust.
